来源:牛血浆 性状(以下信息仅供参考):白色或类白色无定形粉末或固体。 稳定性:水溶液室温下8h内失活。遇热、稀酸、碱、金属等活力降低。 介绍:由动物血浆分离制得的凝血酶原,再用凝血致活酶和氯化钙激活而成。是一种蛋白水解酶,血浆中的可溶性纤维蛋白原在凝血酶的激活下,转变成不溶性纤维蛋白网状结构,使血液凝固。纤维蛋白原有六条肽链组成,凝血酶的组成是从四条肽链的N端切断一定特定的肽键(—Arg—Gly),放出两个A肽(19肽)和两个B肽(21肽),另二条N端为酷氨酸的肽链没有变化。A肽和B肽切除后,减少了蛋白质分子的负电荷,促进了纤维蛋白分子的直线聚合和侧向聚合,从而形成网状结构的纤维蛋白. 注意事项: 1、0.9%的nacl 保存时浓度要大于500U/ml;保存时分装-20℃保存,不能反复冻融,复溶先以放到4℃缓冲1-2h,再拿到常温使用,稀释过程可以直接使用20mMTris-HCl 缓冲液,含 150mM,氯化钠,pH8.0,也可以参考文献按照文献方案。(溶液状态储存,活力会有所下降,酶属于蛋白类,无法避免,如果遇到酶活降低情况,可以降低稀释倍数来达到之前的活性。) 2、纤维蛋白原(约2mg/ml)0.9ml,加凝血酶(10单位/ml)0.1ml,37℃凝固时间大约30秒;纤维蛋白原(约2mg/ml)0.9ml,加凝血酶(5单位/ml)0.1ml,37℃凝固时间大约50秒;溶液均用0.9%氯化钠制备。  3、1ku一支产品总质量大约30mg左右,里面有酶蛋白,缓冲盐和辅料等,单独取用有可能会不均匀,建议整瓶溶解。.  4、酶切条件:融合蛋白1mg,加凝血酶1~5U(如果样品浓度低,体系较大需要增加量),20-37℃孵育16小时左右。有效的消化缓冲液是 20mMTris-HCl 缓冲液,含 150mM,氯化钠,pH8.0。仅供参考
