SDS说明:生化研究,用于从蛋白质中分离核酸,从宿主细胞中脱落某些病毒;蛋白电泳试剂,也可用于质粒提取
SDS生物学应用:
(1)SDS是阴离子表面活性剂常用于蛋白质和类脂类的电泳分离。当其与蛋白质混合,质量比达到1.4:1时,SDS能破坏蛋白质分子间以及其他物质分子间的非共价键使蛋白质的构象发生变化继而使蛋白质变性解离成单一亚基,从而降低或消除了各种蛋白质分子间的天然电荷差异。电泳常用10%的SDS作为储备液。
(2)SDS可以迅速破坏组织结构,抑制RNase和脱氧核糖核苷酶(DNase)活性,所以SDS也是核酸纯化试剂的关键组分。通常使用10%或者20%的SDS储备液,SDS的工作浓度是0.1-0.5%。
SDS贮存条件:密封阴凉干燥保存;
SDS毒性:低毒,大鼠经口半数致死量为1288mg/kg。有刺激性。
